Messenger RNA (mRNA) plays a crucial role in the central dogma of molecular biology as the intermediary between DNA and protein synthesis. Transcribed from DNA by RNA polymerase through a sophisticated process known as transcription, mRNA serves as the blueprint for protein synthesis. During this pivotal process, RNA polymerase binds to the promoter region of DNA, forming a transcription initiation bubble. This bubble allows nucleotides that are complementary to the DNA template to be added in the 5′-to-3′ direction, ultimately producing the functional mRNA transcript required for protein production.

In laboratory settings, this process can be emulated through in vitro transcription (IVT), which combines a DNA template, RNA polymerase, and nucleotide triphosphates (NTPs) within a magnesium-containing buffer. The advantages of IVT-generated mRNA are profound. Notably, this synthetic mRNA does not integrate into the genome, mitigating potential carcinogenic risks. Furthermore, a natural degradation pathway exists for mRNA, ensuring its temporary activity, which is integral for controlled protein expression.
